
Measuring 189.70 miles in the Palmetto State, U.S. 301 travels northeast from the Savannah River at Allendale County to the North Carolina state line at Dillon County. Vastly supplanted by Interstate 95 as the main route in South Carolina, all of U.S. 301 from the Georgia line to Santee, with the exception of the Burton's Ferry Bridge, is four lanes wide. Generally paralleling I-95 northeast of Lake Marion, U.S. 301 is mostly a local route with just two lanes.
U.S. 301 heads east from the Savannah River to Allendale, where it overlaps with U.S. 278 for 0.31 miles along Main Street. U.S. 321 converges with U.S. 301 north from Fairfax at the town of Ulmer. The two routes combine for 1.90 miles north into southwestern Bamberg County.
Continuing northeast from U.S. 321, U.S. 301 leads into the county seat of Bamberg, where it joins U.S. 601. U.S. 301/601 overlap 19.42 miles to U.S. 21 Business (Magnolia Street) in the city of Orangeburg. U.S. 301 turns east from there to meet I-26 at Exit 154, U.S. 176 at Oaks Crossroads and U.S. 15 southwest of Santee.
U.S. 15/301 combine with Interstate 95 north across Lake Marion, and then continue paralleling the freeway to Summerton. U.S. 15 shifts north to Paxville and Sumter while U.S. 301 angles northeast across I-95 again to the Clarendon County seat of Manning.
U.S. 301/521 combine for 1.32 miles along a four lane stretch leading north out of Manning. U.S. 301 continues from there to Sardinia, a 0.72 mile long overlap with U.S. 378 at Turbeville and Olanta in Florence County.
Advancing into Florence, U.S. 301 combines with U.S. 52 for 5.85 miles along a four lane highway. U.S. 301 formerly continued north to Downtown Florence alongside U.S. 52 (Irby Street) and then east on U.S. 76 (Palmetto Street). A bypass for U.S. 301 opened by 2007 around the east side of the city and Florence Regional Airport. Approved by the American Association of State Highway and Transportation Officials (AASHTO) on May 4, 2007, the realignment of U.S. 301 ties into U.S. 76 opposite S.C. 327 (Williston Road), which continues the bypass north to I-95 at Exit 170.
U.S. 301 overlaps with U.S. 76 east from S.C. 327, 8.83 miles to Pee Dee in western Marion County. U.S. 76 leads to Marion and Mullins while U.S. 301 runs northeast to Sellers and U.S. 501 at Latta. U.S. 501 joins U.S. 301 for 14.28 miles to Dillon and the state line at South of the Border.
