A rural route for the most part, SR 29 travels 75.82 miles from U.S. 27 near Palmdale in Glades County to U.S. 41 at Carnestown in southern Collier County. The state road joins the city of Immokalee with I-75 (Alligator Alley) to the south and the Hendry County seat of La Belle to the north. Collier County Road 29 extends the roadway south from SR 29 and U.S. 41 another 7.75 miles to Everglades City and Chokoloskee.
