There are two sections of County Road 427 in Central Florida. The southern branch follows Maitland Avenue 1.328 miles north from U.S. 17/92 at Downtown Maitland to the Seminole County line, and another 1.571 miles north to SR 436 (Semoran Boulevard). This section is mostly unmarked on the Orange County portion. 0.478 miles along SR 436 separate the two legs of CR 427 along SR 436.

North from Semoran Boulevard, CR 427 travels another 10.435 miles, mostly paralleling U.S. 17 & 92. Following Ronald Reagan Boulevard, a four-lane divided parkway, CR 427 passes through Altamonte Springs and Longwood to southern reaches of Sanford. Ronald Reagan Boulevard transitions to Sanford Avenue north of the Central Florida GreeneWay (SR 417). CR 427 ends there, with CR 425 taking over northward 1.62 miles to SR 46 (East 25th Street) in Sanford.

Ronald Reagan Boulevard crosses a CSX Railroad line at-grade and next intersects General Hutchinson Parkway east through Spring Hammock Preserve to U.S. 17/92. 09/30/17
CR 427 encircles the northern boundary of Spring Hammock Preserve from General Hutchinson Parkway to CR 15 (Country Club Road). Big Tree Crossing business park lines the west side of the parkway on the ensuing stretch. 09/30/17

The first shield for County Road 427 follows the transition from Sanford Avenue (CR 425) south onto Ronald Reagan Boulevard. 03/21/08
Ronald Reagan Boulevard quickly intersects Lake Mary Boulevard beyond Sanford Avenue. The east-west arterial stretches across most of Sanford, linking the city with Lake Mary and Orlando-Sanford International Airport (SFB). 03/21/08

SR 434 comprises a major arterial route from Longwood west to I-4 and Altamonte Springs and east to Downtown Winter Garden. The state route encircles the Orlando metro area southwest to Forest City and southeast to the University of Central Florida (UCF). 03/06/08
Ronald Reagan Boulevard heads south from SR 434, paralleling SunRail to the east by an area of warehouses and Lyman High School. 03/06/08
Dog Track Road is an arterial route connecting Ronald Reagan Boulevard with U.S. 17/92 at Seminola Boulevard south into Casselberry. The road derives its name from Sanford Orlando Kennel Club, located nearby. 03/06/08
